Introduction

• Non- narcotic analgesic removed from over the counter remedies in USA because of adverse effects:

Pharmacokinetics

• metabolized to acetaminophen
  

• elimination via liver
• 1/2life = 0.6 hours
• protein binding = 39 %

Adverse-effects

• nephrotoxicity (including capillary necrosis)
• hemolysis
• thrombocytopenia
• methemoglobinemia

Laboratory

• specimen: plasma ( EDTA)
• methods: GLC, HPLC, color, GC-MS
• interferences:

• theophylline interferes with some HPLC procedures
• silyl derivatives for GLC are unstable
• colorimetric assays are non-specific
